[Abstract]

Objective: To investigate the inhibition effects of ?-ANO and ?-ANO monomer on human prostate cancer.Method: The monomers of ?-ANO and ?-ANO were purified through alumina column chromatographic separation and detected through lamellar analytic process respectively. SRB staining was applied to investigate the suppression effect of ?-ANO and ?-ANO on androgen-dependent and androgen-independent cells. Cell morphage was examined under microscopy. ELISA method was used to examine the effect of ?-ANO on the secretion of PSA by LNCAP cell line. Athymic nude mouse solid tumor model, subcutaneously inoculated human prostate cancer cells, was used to investigated the suppression effect of the drug to the proliferation of the tumor.Result: 1. Both ?-ANO and ?-ANO monomers effectively inhibited human prostate cancer cell proliferation, and the suppression effect of ?-monomer was stronger than that of ?-monomer. Both high concentrations of ?-ANO and ?-ANO could perform cell killing effects and significantly change cytomorphology on different human prostate cancer cell lines. In the androgen-depleted medium, ?-ANO could stimulate the growth of LNCaP and L1A in a concentration dependent manner at the range of 0.1-1?mol/L, but ?-ANO at high concentration(>1?mol/L) inhibited the proliferation. In the medium with R1881 (a kind of androgen stimulant), both ?-ANO and ?-ANO could antagonized the proliferation effect of R1881 while the effect of ?-ANO was more weaker. The secretion of PSA was decreased by ?-ANO pretreated LNCaP cells, and the suppression effect was more significant in the presence of R1881. ?-ANO could effectively suppress the growth of human prostate cancer RV1 implanted to nude mouse.Conclusion: Both ?-ANO and ?-ANO have strong inhibiting effects on prostate cancer in vitro, and the suppression effect of ?-ANO is stronger than that of ?-ANO.
